p38 MAPK蛋白在米诺环素抑制小鼠EAE发病中的表达变化及意义 被引量:1

摘要 【目的】探讨米诺环素抑制小鼠实验性变态反应性脑脊髓炎(EAE)发病过程中,p38 MAPK蛋白的表达变化及意义。【方法】将小鼠随机分为对照组、EAE模型组和米诺环素治疗组。在动物发病高峰期取小鼠脑组织和脊髓,行HE组织染色观察病理改变,以及Western Blot方法检测p-p38 MAPK蛋白的表达。【结果】①EAE组脑组织和脊髓有多部位的血管炎,血管周围大量炎性细胞浸润,小血管明显扩张,改变以白质为著。治疗组的血管炎范围、数量及程度均明显减轻。②p-p38 MAPK蛋白在对照组仅有少量表达;在EAE组表达明显升高,达3倍左右;治疗组的表达显著降低,但仍较对照组为高。【结论】p38 MAPK途径可能参与了米诺环素对小鼠EAE发病过程的抑制作用。 [Objective]To investigate the dynamic expression and the role of p38 MAPK in brain and spinal tissue of experimental allergic encephalomyelitis(EAE) mice treated with minocycline. [Methods]C57BL/6 mice were randomly "divided into the control group, the EAE model group and the minocycline treatment group. At peak stage of disease, the brain and spinal tissue were stained with HE for pathologic investigation, and were detected for p-p38 MAPK with Western Blot method. [Results](1)Mice with EAE were found with obvious pathological changes. HE staining showed vasculitis throughout central nervous system (CNS), vascular dilatation and perivascular infiltration of inflammatory cells. Lesions in minocycline treatment group were less frequent and markedly reduced destructions. (2)The expression of p-p38 MAPK protein was a little in control group, but significantly elevated to 3 times in EAE group and markedly lower in minocycline group which was still higher than the level of the control group. [Conclusion]P38 MAPK pathway might be a mechanism for the inhibition of EAE by minocycline.
