摘要 目的探讨紫杉醇诱导非小细胞肺癌细胞的死亡形式及B i m的作用。方法用M T T法检测细胞死亡;AnnexinⅤ染色流式细胞术检测细胞凋亡;Western blot检测紫杉醇诱导前后Bim蛋白的表达水平。结果紫杉醇诱导肺癌细胞死亡具有剂量依赖性;A549细胞的死亡形式主要为细胞凋亡;Bim蛋白表达水平在紫杉醇诱导的非小细胞肺癌A549中显著升高。结论紫杉醇通过诱导细胞凋亡杀伤非小细胞肺癌,细胞凋亡可能与Bim表达上调有关。 Objective To investigate the forms of paclitaxel-induced cell death of non-small-cell lung cancer cell lines (NSCLC) and the role of Bim. Methods Cell viability was quantitated by MTT assay. Apoptosis was measured by FITC-Annexin V flow cytometry. The expression of Bim at protein levels before and after paclitaxel treatment was measured by Western blot analysis. Results Paclitaxel induced apoptosis of NSCLC in a dose- dependent manner. Apoptosis was the main form of cell death induced by paclitaxel in A549 cells. The expression of Bim at protein levels was significantly increased in A549 cells after exposure to paclitaxel. Condusion Paclitaxel kills A549 cells by induction of apoptosis, which may be associated with the up-regulation of Bim.
