摘要
目的阐明猪血红蛋白的主要理化特性,并了解戊二醛聚合反应对猪血红蛋白主要性能的影响。方法猪血经多步纯化获取高纯度猪血红蛋白,以戊二醛为交联剂制备聚合猪血红蛋白样品。鉴定猪血红蛋白的纯度、聚合体分子量分布、携氧性能及自氧化等指标。结果猪血红蛋白在生理pH条件下的P50为23.6 mmHg,Hill系数为2.86。聚合猪血红蛋白为分子量大小不均一的混合体,聚合体P50和Hill系数均减小,自氧化趋势增大。结论猪血红蛋白具有较高氧亲和力,与人血红蛋白相似,明显高于牛血红蛋白。猪血红蛋白的丰富来源、特殊的氧亲和力和较好的化学稳定性等特点使其可成为一种血红蛋白类氧载体产品制备的有用材料。
Aim This study was to demonstrate the major physical and chemical properties of porcine hemoglobin (pHb) and polymerized porcine hemoglobin. Methods Ultra-pure pHb was obtained through multiple purification steps; with glutaraldehyde as the cross-linker, pPolyHb was then produced; purity of the purified pHb, molecular weight distribution of pPolyHb, oxygen affinity and auto-oxidation of Hb were analyzed. Results At a physiologi- cal pH condition, pHb had a P50 about 23.6 mmHg and Hill coefficient about 2.86. pPolyHb was composed of Hb polymers of various molecular sizes with increased oxygen affinity and decreased Hill coefficient. The polymerization increased the tendency of auto-oxidation of the products. Conclusion pHb has high oxygen affinity, similar to that of human Hb and much higher than that of bovineHb. Rich supply, the unique properties of P50 of pHb could make it a useful material in hemoglobin-based oxygen carriers(HBOCs).
