石油降解菌的筛选、鉴定及降解石油的初步研究 被引量:4

摘要 从被石油污染的土壤中分离得到一株石油降解菌O-D-1,通过形态、生理生化实验和16S rDNA序列分析对其进行了鉴定。结果表明,该菌的形态及生理特征与芽孢杆菌相似,芽孢杆菌的代表菌株B.anthracis、B.cereus、B.thuringiensis与该菌的同源性达99%。从系统发育树也可以看出,该菌与苏云金芽孢杆菌B.thuringiensis的距离最近,归属于芽孢杆菌属(Bacillus)。通过对降解条件的研究,确定了降解菌O-D-1的最适生长条件为:初始pH值5、温度37℃、渗透压1%NaCl、辅助营养物质蛋白胨浓度4 g.L-1。 An oil-degradation bacterium from oil contaminated soil is screened and identified by the physiological and biochemical tests and its 16S rDNA sequencing. The isolated bacterium is similar to Bacillus on physiological and biochemical phenotype. The sequence of its 16S rDNA indicates that there is 99% homology to the representative strains of B. anthracis, B. cereus and B. tburingiensis. And in the development tree this strain is the nearest to B. tburingiensis. Through the oil-degradation study of the strain, the optimized culture conditions for its growth are as follows, original pH value is 5, culture temperature is 37℃, osmotality is 1% NaCl,concentration of peptone is 4 g ·L^-1.
