摘要
目的:以柴胡中柴胡皂苷a及柴胡皂苷d含量为评价指标,优选柴胡最佳提取工艺。方法:采用L9(34)正交试验设计对提取方法进行优选,HPLC-ELSD法测定柴胡皂苷a及柴胡皂苷d含量。采用Agilent TC-C18(4.6mm×250mm,5μm)色谱柱,流动相为乙腈-水(50∶50);流速:0.8mL·in-1;柱温:20℃;ELSD为检测器。结果:最佳提取方法为回流1h,加5%氨水甲醇30mL。结论:优选出的提取方法经方法学考察符合要求,可为柴胡中化学成分的提取提供参考,使分析结果准确、可靠。
OBJECTIVE To explore the optimum extracting conditions for Radix Bupleuri. METHODS The orthogonal design was taken for the optimization, the content of Saikosaponin a and Saikosaponin d was determined by HPLC-ELSD with Agilent TC-C18 (4. 6 mm× 250 mm, 5 μm)column; mobile phase was Acetonitrile-H20(50: 50) ; the flow rate was 0. 8 mL· min^-1 ; the column temperature was 20 ℃ ; it was determined by ELSD. RESULTS The optimum extracting conditions for Radix Bupleuri were as follows:circumfluence lhours,extracting with 5% hartshorn- MeOH 30 mL. CONCLUSION The method of the optimum extracting procedure is well, The method can be used in the extracting of saikosaponin in Radix Bupleuri. The result is reasonable and stable.
