STAT3在围植入期小鼠子宫内膜的表达及意义

摘要 目的:检测信号转导和转录激活因子3(STAT3)在小鼠围植入期子宫内膜的表达,探讨STAT3在胚泡植入中的生物学作用。方法:取未孕动情期、真孕及假孕1天、2天、3天、4天上午、4天下午、4天午夜、5天、6天、7天、8天小鼠子宫做切片,用免疫组化技术和图像分析方法对STAT3在围植入期子宫内膜的表达进行定位和半定量分析。结果:STAT3主要分布于小鼠子宫内膜腔上皮、腺上皮和蜕膜细胞。真孕组小鼠子宫内膜腔上皮和腺上皮的STAT3于孕4天上午开始呈强阳性表达,孕4天下午表达最强,此后表达逐渐减弱;蜕膜细胞的STAT3表达则从孕5天开始逐渐增强。假孕组小鼠子宫内膜中STAT3只在孕4天呈弱阳性表达。未孕组小鼠子宫内膜中STAT3弱表达。真孕组小鼠子宫内膜中STAT3含量明显高于同期各假孕组。结论:STAT3参与胚泡植入过程和子宫内膜容受性的建立。STAT3还可能参与植入后胚泡滋养层的扩展、分化及子宫内膜蜕膜化反应。STAT3在子宫内膜的表达不仅受母体因素调控,而且可能与胚泡的刺激有关。 Objective:To observe the expression of signal transducer and activator of transcription 3 (STAT3) in the peri-implantation mice endometrium and to study the biological effects of STAT3 in the process of blastocyst implantation. Methods:Uteri of mice in the non- pregnant ( estrous stage), pregnant and pseudopregnant state of 1 - 8 days were obtained. Im- munohistochemistry was used to observe and analyze the expression and distribution of STAT3 in the uterine endometrium during mice embryo implantation. An image analysis was also per- formed. Results:STAT3-specific staining was mainly distributed in the luminal epithelium, glan- dular epithelium and decidual cells. In the pregnant group, the expression of STAT3 was stron- ger positive in the luminal and glandular endometrium on the 4th day morning, peak at 4th day afternoon,then declined, but increased in decidual cells. In the pseudopregnant group, weak staining of STAT3 was only observed on the 4th day of gestation ,and negative staining were observed on the other days. STAT3 was expressed weakly in the endometrium of nonpregnant mice. The expression level of STAT3 in mice endometrium of the pregnant group was much higher than that in the homeochronous pseudopregnant group. Conclusions:STAT3 may contribute in blastocyst implantation and the establishment of uterine receptivity. STAT3 also may indirectly take participate in the process of trophoblast outgrowth, differentiation and decidualization of endometrium. The expression of STAT3 in mice endometrium is regulated not only by materhal factors (steroid hormones), but also by the implanted embryos.
