黄芪牛蒡子不同配伍对糖尿病大鼠肾组织结缔组织生长因子的影响 被引量:1

Effects of Different Compatibility of Astragalus and Arctium on the Expressions of Connective Tissue Growth Factor in Kidney of Diabetic Rats

摘要 目的探讨黄芪与牛蒡子不同配伍对糖尿病大鼠肾组织病理学及结缔组织生长因子(CTGF)表达的影响。方法建立链脲佐菌素(STZ)大鼠模型,将黄芪、牛蒡子各分为高中低剂量,按照正交设计进行不同剂量的组合。大鼠灌胃4周或8周,分别检测尿蛋白及肾组织病理学改变,RT-PCR法检测大鼠肾组织内CTGF的基因表达。结果黄芪与牛蒡子配伍可减少STZ模型大鼠24h尿蛋白的排出,抑制系膜基质的增加,下调肾组织CTGF mRNA的过高表达。实验第4周时,以黄芪中低剂量为主的配伍作用效果较好;第8周时,以黄芪高剂量为主的配伍在减少24h尿蛋白、抑制系膜基质积聚和降低CTGF mRNA的表达方面作用明显,同时牛蒡子的剂量不宜过大。结论黄芪与牛蒡子配伍可降低肾组织CTGF的表达,减轻糖尿病肾脏病变;在病变不同阶段,黄芪与牛蒡子配伍剂量宜进行调整。 Objective To investigate the expressions of different compatibility of Astragalus and Arctium on connective tissue growth factor (CTGF) in renal tissue of diabetic rats. Method Streptozotocin(STZ)-induced diabetic rats were established as experimental model. Astragalas and Arctium were divided into three dosages (low, moderate, high)respectively and combined according to orthogonal experimental design and were administered to the diabetic rats. On the 4th and 8th weekends, HE and PAS stainings were used to observe the changes of glomerular ECM and CTGF mRNA expression was analyzed by reverse transcrlption-polymerase chain reaction (RT-PCR). Results Compatibility of Astragalus and Arctium could reduce 24h urinary protein and suppress mesenteric matrix in diabetic rats, with better results by low- and moderate-dose of Astraga/as in the 4th week and by high-dose in the 8th week, but Arctium should not be over- dosed. Conclusion The compatibility of Astragalas and Arctium may ameliorate the renal lesion of DN by decreasing the expression of CTGF, and the doses of Astragalus and Arctium should vary with pathological phases.
