摘要
目前,在水稻中已克隆出GA受体GID1,其功能丧失会导致植株矮化。同时,在拟南芥中也克隆出三个相关的GA受体。通过它们氨基酸序列的保守区设计兼并引物,然后用上述引物在玉米中进行扩增。扩增出的基因片断大小约为700bp,然后进行产物的回收及测序,根据这个片段从数据库中得到对应EST序列,其中包括一小段5’、3’侧翼序列以及起始密码子在内的全部翻译序列。研究发现水稻与玉米GA受体的氨基酸序列具有较高的同源性,达到80.5%。由此推断,此基因可能就是GA受体在玉米中的同源基因。
At present, GID1 as gibberellin acid ( GA ) acceptor has been cloned in rice , loss of its function can cause dwarfing of plant height. At the same time, three GA acceptors have been cloned in Arabidopsis. According to their conserved domains, pairs of degenerate primers were designed. Amplifications were carried out in maize with those pairs of degenerate primers. A fragment with a size of 700 base pairs was obtained in maize. Subsequently, this fragment was purified and sequenced, the sequence was compared with a software to find its corresponding EST sequence. Results revealed that an EST, including 5 and 3' flanking sequences as well as an entire coding frame was found . According to the EST, the amino acid coding this gene was deduced . Their amino acid had higher similarity with 80.5% in rice and maize. It could be deduced that this gene may be an homologous gene of GA acceptor in maize.
