视瞻昏渺发病机理与VEGF、bFGF、PEDF表达 被引量:2

摘要 目的:观察脉络膜新生血管(CNV)动物模型视网膜色素上皮至脉络膜之间细胞因子VEGF、bFGF、PEDF表达的变化。方法:用半导体激光光凝建立CNV动物模型,于造模后1w,2w,3w,4w,8w分别行免疫荧光检查(FFA);于造模后8w处死动物行HE染色观察视网膜和脉络膜的病理组织学变化,免疫组化法检测VEGF、bFGF、PEDF表达。结果:HE染色可见脉络膜新生血管形成。造模后模型组VEGF、bFGF上调,VEGF/PEDF值升高,与正常对照组比较,差异有统计学意义(P<0.01);PEDF值轻度上调,与正常对照组比较差异无统计学意义(P>0.05)。结论:半导体激光可诱导CNV动物模型,该模型可做为视瞻昏渺的动物模型进行实验研究。 Objective:To observe the changes of VEGF,bFGF and PEDF expressions in retina and choroidal of choroidal neovascularization(CNV) rabbit models.Methods:6 rabbits in model group were photocoagulated by semiconductor laser in eyes.On the first, second,third ,fourth, eighth week after photocoagulation, histological changes of retina and choreidal were observed by fundus fluorescein angiography(FFA)examination;the changes of VEGF, bFGF and PEDF expressions in retina and choroidal of CNV rabbit models were detected by immunohistochemistry.Results:On the second week after model establishment, disciform leakage staining appeared in the FFA;on the eighth week after model estabhshment, compared with the control group,the expressions of VEGF,bFGF and VEGF/PEDF were significantly increased(P〈0.01 ),PEDF expression was lightly increased (P〉0.05)in model group.Conclusion :CNV rabbit model induced by semiconductor laser can be a model for experimental study of Shizhan exudative senile macular degeneration(Shizhan Hunmiao).
