补虚化瘀祛痰饮对老年冠心病的抗氧化作用机制研究 被引量:2

Antioxidant Mechanism of Buxu Huayu Qutan Drink in Treating Senile Coronary Heart Disease

摘要 目的:在论证老年冠心病中西医基本病机的基础上,研究补虚化瘀祛痰饮组方构成的抗氧化作用机制。方法:将66例老年冠心病患者随机分为中药干预组和对照组,2组常规治疗方法相同,中药干预组另加用补虚化瘀祛痰饮。采用生化及分子生物学手段测定治疗前、后超氧化物歧化酶(SOD)活性、血浆脂质过氧化物(LPO)含量以及外周血单个核细胞MnSOD基因mRNA的表达水平。结果:对照组患者治疗前、后血浆SOD活性及LPO含量无显著性差异(P>0.05),中药干预组患者治疗后血浆SOD活性显著升高,LPO含量显著降低(P<0.01);中药干预组MnSOD基因mRNA表达水平显著高于对照组(P<0.01)。结论:补虚化瘀祛痰饮能诱导单个核细胞MnSOD基因表达,干预氧自由基的产生,并增强消除氧自由基的功能。 OBJECTIVE:To study the antioxidant mechanism of Buxu huayu qutan drink in treating senile coronary heart disease by justifying the basic pathogenesis of senile coronary heart disease in both Chinese medicine and western medicine. METHODS: A total of 66 patients with senile coronary heart disease were randomly assigned to receive conventional therapy alone(control group)or conventional therapy in combination with Buxu huayu qutan drink, a traditional Chinese medicine(trial group) .The SOD(superoxide dismutase) activities, serum lipid peroxidation(LPO) level and the mRNA expression level of MnSOD gene in mononuclear cells in peripheral blood in two groups were determined before and after treatment by using the biochemical and the molecular biology techniques. RESULTS:As compared with before treatment, there was no significant difference in the control group after treatment regarding the activity of SOD and the levels of LPO(P 〉0.05).However,in the treatment group, the activity of SOD increased significantly and the level of LPO decreased markedly after treatment(P 〈 0.01).The results indicated the mRNA expression of mononuclear cell MnSOD gene in trial group was significantly higher than in the control group (P 〈 0.01) .CONCLUSION:Buxu huayu qutan drink can induce the expression of MnSOD gene of mononuclear cells,interfere in the production of free radical and promote the elimination of oxygen free radicals.
