摘要
目的观察抗转化生长因子-β2抗体(抗TGF-β2抗体)对体外培养的滤过泡成纤维细胞(FB)周期的影响。方法利用手术修复青光眼小梁切除术后失败滤过泡时切除下来的瘢痕组织进行原代细胞培养,获得FB。将浓度为0、0.01、0.1、1mg/L的抗TGF-β2抗体作用于FB 24h以及用半数抑制率(IC50)0.14mg/L作用0、24、48、72h,通过流式细胞仪(FCM)检测抗TGF-β2抗体对细胞周期的影响。结果随着抗TGF-β2抗体浓度增大及作用时间的延长,FBG1期细胞百分数增多,而S、G2期的细胞含量逐渐减少,FB增殖指数呈下降趋势(P<0.05)。结论抗TGF-β2抗体能阻断FB从G1进入S期,使细胞停滞在G1期,从而抑制细胞的增殖过程。
Objective To observe the role of anti-TGF-β2 antibody in the cell cycle of cultured fibroblasts in filtering bleb. Methods Fibroblasts were isolated from the filtering bleb after trabeculectomy in patients with glancoma. The primary cells were treated with anti-TGF-β2 antibody at different concentrations (0, 0.01, 0. 1 and 1 μg/ml) for 24 hours and at IC50(0.14 μg/ml) for 0, 24, 48 and 72 hours, and then their cell cycles were determined by flow cytometry. Results Percentage of GI cells increased, but that of S and G2 cells and proliferation index decreased with the concentration and action time of anti-TGF-β2 antibody (P〈0.05).Conclusion Anti-TGF-β2 antibody may inhibit fibroblast proliferation by arresting G1 ceils.
