摘要
[目的]建立白癜风胶囊中补骨脂素和异补骨脂素含量测定方法。[方法]采用单因素与正交试验设计法考察最佳提取条件,采用高效液相色谱法测定,色谱柱:Supelcosi LC18柱(5Ⅲ,4.6mm×250.0mm);流动相:甲醇-水(40:60),流速:1.0ml/min;柱温:35℃;检测波长:245nm。[结果]补骨脂素在5.5~55.0μg/ml(R^2=0.9999,n=8),异补骨脂素在4-8—48.0μg/ml(R^2=0.9999,n=8)范围内线性关系良好。补骨脂素的平均加样回收率为100.1%,RSD=1.1%(n=6);异补骨脂素的平均加样回收率为99.9%,RSD=1.6%(n=6)。[结论]方法简便、准确、可行,可有效控制白癜风胶囊的质量。
[Objective] The research aimed to establish a kind of method for determining the content of psomlen and isopsoralen in vitiligo capsule. [ Method] The optimum extraction conditions were investigated by using the design of single factor and orthogonal test. The content of psomien and isopsomien in vitiligo capsule were detemined by using high performance liquid chromatography (HPLC) under the conditions including taking Supelcosi LC18 column ( 5μm, 4.6 mm × 250.0 mm) as chromatographic column, with methanol-water( 40: 60 ) as mobile phase, the flow rate of 1.0 ml/rain, the column temperature at 35℃, the detection wavelength at 245 nm. [ Result ] A good linearity was seen in the calibration curves when the detection concentration of psomien and isopsomien were 5.5 - 55.0 μg/ml ( R^2 = 0.999 9, n = 8 ) and 4.8 - 48.0/q/ml ( R2 = 0.999 9, n = 8) resp. The average addition recovery of psomien was 100.1% ,with RSD of 1.1%(n =6). The average addition recovery of isopsomien was 99.9%,with RSD of 1.6%(n =6). [Conclusion] This method is simple, accurate and feasible and the quality of vitiligo capsule could be effectively controlled.
