摘要
目的观察GnRH受体在人卵巢癌细胞系SKOV3、OVCAR3中的表达,并研究GnRH激动剂(Triptorelin)对卵巢癌细胞生长的抑制作用。方法采用免疫组化SP法检测两种细胞系内GnRH受体的表达,分别应用不同浓度Triptorelin作用于细胞,MTT法检测细胞生长抑制率。结果人卵巢癌细胞系OVCAR3表达GnRHⅠ受体,SKOV3细胞系不表达GnRHⅠ受体,Trip-torelin可明显抑制GnRHⅠ受体阳性的OVCAR3细胞生长,对GnRHⅠ受体阴性的SKOV3细胞生长无明显抑制作用。结论GnRH激动剂Triptorelin通过与人卵巢癌细胞株GnRHⅠ受体结合,发挥抗增殖的作用。
Objective To observe the expression of gonadotropin releasing hormone receptor in SKOV3,OVCAR3 ovarian cancer cell lines,and investigate the inhibitory action of GnRH agonist. Methods Immunohistochemical SP was used to detect the GnRH receptor on SKOV3 and OVCAR3 cell lines.Different concentration of GnRH agonist were used to SKOV3 and OVCAR3 cell lines.The cells inhibition rates were detected by MTT technique. Results OVCAR cells show GnRH Ⅰ receptor immunoreactivity and triptorelin have growth inhibition action on these cells;but SKOV3 ceils didn't show GnRH Ⅰ receptor immunoreactivity,and triptorelin have no inhibiton action on these cells. Conclusion In ovarian cancer cell lines, the inhibitory effect of GnRH agonist was caused by it's combination to GnRH Ⅰ receptor.
