海洋小单孢菌来源的轻快菌素B FW523-3的体外抗肿瘤活性 被引量:12

摘要 目的考察轻快菌素B FW523-3的体外抗肿瘤活性。方法采用MTT比色分析法、流式细胞法和高内涵药物筛选的方法检测FW523-3对肿瘤细胞株细胞生长、细胞周期、肿瘤细胞骨架蛋白、线粒体膜电位的影响。结果FW523-3对肿瘤细胞株K562、L929的生长有明显抑制作用,IC50分别为500和400ng/ml。FW523-3处理后的K562肿瘤细胞阻滞在G0/G1期。FW523-3使L929肿瘤细胞的细胞核大小、细胞骨架纤维状肌动蛋白F-Actin量、线粒体膜电位明显减少。结论FW523-3有显著抗肿瘤活性。 Objective To investigate the antitumor activities of rakicidin B FW523-3 in vitro. Meth ods The inhibitory effects of the FW523-3 to the proliferation, cycle, skeleton, mitochondrial membrane potential of the tumor cells were assayed by the colorimetric MTT assay, flow cytometry and high content screening methods. Results FW523-3 strongly inhibited the cell proliferation of K562 and L929 tumor cell lines with IC50 value of 500ng/ml and 400ng/ml, respectively. K562 cells were arrested in G0/G1 phase when treated with FW523-3, which had the ability to decrease the size of nucleolus, disrupt tumor cell F-actin synthesis and reduce mitochondrial membrane potential. Conclusion FW523-3 exhibited potent antitumor activities.
