摘要
目的:研究潜阳熄风、化痰通腑、益气活血3法对脑缺血再灌注损伤大鼠神经细胞凋亡的影响。方法:采用阻断大鼠大脑中动脉再灌注方法复制局灶性脑缺血再灌注模型,用流式细胞术和TUNEL法,分别观察脑缺血半暗带神经细胞凋亡率和阳性染色细胞的变化。结果:脑缺血再灌注模型大鼠缺血半暗带神经细胞凋亡是一个动态过程,1天达最高峰,以后逐渐减弱;与模型组比较,各治疗组神经细胞凋亡率显著降低,阳性染色细胞显著减少;各治疗组间比较,1天无显著差异,3天、7天益气活血法优于潜阳熄风、化痰通腑法。结论:3种中医治法均有抗脑缺血再灌注损伤大鼠缺血半暗带神经细胞凋亡的作用,对神经细胞有保护功能。
Objective:To observe the effects of three kinds of Chinse therapeutic methods:suppressing the hyperfuction of the liver-yang and stopping endogenous wind to relieve convulsion,resolving phlegm and purgation,supplementing qi and activating blood circulation,on neurocyte apoptosis of rats with cerebral ischemia-reperfusion.Methods: The models were made by blocking the middle cerebral arteries of rats.The neurocyte apoptosis in the ischemic penumbral area was detected by flow cytometry and TUNEL respectively.Results:The neurocyte apoptosis in the ischemic penumbral area got the highest on the first day,than it decreased gradually;the neurocyte apoptosis rate of each treated group was obviously lower than that in the model,positive cells of staining reduced significantly.The effects of supplementing qi and activating blood circulation were better than the other two methods on the third and seventh day.Conclusion: Three kinds of Chinse therapeutic methods can prevent the neurocyte apoptosis of rats with cerebral ischemia-reperfusion.
