摘要
目的本文观察了一氧化氮供体型沙利度胺衍生物WⅠ5对培养人脐静脉内皮细胞(ECV-304细胞)分泌一氧化氮以及形态学的影响。方法用不同浓度的沙利度胺和WⅠ5作用于ECV-304细胞,采用NO测试盒测定细胞培养上清液的NO含量,间接反映细胞的NO生成量。结果随着沙利度胺浓度增高,ECV-304细胞NO释放量逐渐减少,高浓度时NO释放量明显减少(P<0.05);而随着WⅠ5浓度增高,ECV-304细胞NO释放量逐渐增加,高浓度时NO释放量极显著性增加(P<0.01)。结论显示一氧化氮具有促进细胞增殖和抑制细胞增殖的双重特性,其促进增殖和抑制增殖的作用很大程度上依赖于NO释放量的多少。
OBJECTIVE To investigate the effects of thalidomide and NO donating-thalidomide derivative on NO production in ECV-304 cells. METHODS NO test box reaction was used to detect respectively the NO-content in supernatant of cultured cells with effects of thalidomide and NO donating-thalidomide derivative in different concentrations. RESULTS NO production of thalidomide was inhibited in high concentration of thalidomide (P〈0.05), but was increased with the increasing concentration of W I 5 (P 〈 0.01).NO production was increased obviously according to the increasing concentration of W I 5 (P 〈 0.01). CONCLUSIONS The double regulation of NO can increase or inhibit the endothelial cells (ECV-304), the degree of increasing or inhibiting cells proliferation was depended on NO production.
