摘要
目的:观察原癌基因c-met在不同分化的胃癌细胞株中的表达水平。方法:采用实时荧光定量PCR仪和Westernblotting检测高分化胃癌MKN28、中分化胃癌SGC-7901、低分化胃癌MKN45和正常胃细胞GES1中c-met的表达情况。结果:荧光定量PCR显示原癌基因c-met在三个胃癌细胞株中均有高表达,但以中分化的胃癌细胞SGC-7901的表达(0.064606±0.0025)显著高于低分化的胃癌细胞株MKN45(0.016657±0.0002)和高分化的胃癌细胞株MKN28(0.002419±0.0002)(P<0.01);Westernblotting显示c-met在蛋白水平的结果与前一致。结论:四个胃癌细胞株中,原癌基因c-met均有表达,以SGC-7901表达量最高。
Objective: To investigate the expression of proto-oncogene c-met in different differentiated gastric cancer cell lines. Methods:Expression of c-met was detected with SYBR Green I quantitative real-time PCR and Western blotting in 4 gastric cancer cell lines, including well-differentiated MKN28, moderately differentiated SGC- 7901, poorly differentiated MKN45 and normal gastric cell line GES1. Results:Real-time PCR detected high levels of c -met expression in all three gastric cancer cell lines, particularly the SGC-7901 (0.064606±0.0025) as compared with others. Western blotting showed findings of c-met proteins consistent with PCR results (P〈0.01). Conclusion:C-met was expressed in all the gastric cancer cell lines studied, especially in SGC-7901.
