天麻多糖提取与自由基清除作用研究 被引量:10

Extraction and free radicals scavenging activities of polysaccharides from Gastrodia elata B1

摘要 目的:研究活性天麻多糖的提取与自由基清除作用。方法:以水提醇沉法制备天麻粗多糖,CTAB季铵盐络合法、凝胶过滤柱层析进一步纯化;利用Feton体系和邻苯三酚自氧化反应,采用比色法测定天麻多糖对OH·及O2-·自由基的清除作用。结果:通过正交实验获得天麻多糖提取工艺为料水比1∶40、浸提温度80℃、提取次数2次,75%乙醇沉淀;在相同的试验体系浓度下,天麻粗多糖及纯化多糖对OH·的IC50分别为1.18、1.62mg/mL;对O2-·的IC50分别为0.81、1.03mg/mL。结论:天麻多糖对氧自由基具有较佳清除能力。 Objective: To study the Gastrodia elata B1 polysaccharides(GPS) extraction and free radicals scavenging activities. Methods: The polysacchaddes was extracted from Gastrodia elata B1 by water and precipitated by ethanol, pudfied with CTAB and the sepharose 6-fast flow chromatography. In the vitro simulated chemical reaction system, the polysaccharide scavenging effect on OH. with Fenton reaction and O^2-. with Pyrogallol Autoxidation Method was studied. Results: The optimal extraction technology conditions were that the temperature was 80 ℃, the extraction time was twice, the solid-liquid ratio was 1:40 and the precipitating ethanol concentration was 75%. The respective IC50 of raw GPS and refined GPS to OH. was 1.18 mg/mL and 1.62 mg/mL, to O^2-. was 0.81 mg/mL and 1.03 mg/mL. Conclusion: These results clearly indicated that GPS had a good free radicals scavenging activity.
