磁性阳离子纳米脂质体的制备及其性能表征

摘要 目的探讨磁性阳离子纳米脂质体的制备方法及其性能特征。方法采用薄膜分散法制备磁性阳离子纳米脂质体,并且对其形貌、粒径、磁场响应性、细胞毒性进行表征。结果薄膜分散法制备磁性阳离子纳米脂质体方法简单易行,制备的磁性阳离子纳米脂质体的形貌呈球形,平均粒径为98nm;且具有超顺磁性,在水溶液中的磁场响应性能良好;MTT实验表明磁性阳离子纳米脂质体是毒性较小的基因转染载体。结论薄膜分散法制备的磁性阳离子纳米脂质体粒径小、分布均匀、细胞毒性小,将可作为肝癌基因治疗一种理想的非病毒转染载体。 [Objective] To explore preparative method and performance characteristics of nano-magnetie cationic liposomes. [Methods] We prepared nano-magnetic cationic liposomes by film dispersion method .The properties of size, morphology, magnetic field .responsiveness and cell toxicity are studied by some testing means. [Results] The preparative method of nano-magnetie cationic liposomes is very simple and easy. Particle diameter analyzing and TEM indicated that nano-magnetic cationic liposomes present sphericity, the average diameter of particles is about 98 nm. The nano-magnetie cationic liposomes present favorable responsibility to magnetism in water solution and with superparamagnetism. MTr assay showed that nano-magnetic cationic liposomes was a superior gene transfection vector with low toxicity. [Conclusions] Our results showed that nano-magnetie cationic liposomes prepared by film dispersion method were uniform, the diameter and cell toxicity were smaller, It can be used as an ideal non-viral transfection carrier for liver cancer therapy.
