摘要
目的探讨醛固酮(ALD)对大鼠肾小球系膜细胞结缔组织生长因子(CTGF)基因表达的影响。方法①以不同浓度的ALD(10-11、10-10、10-9、10-8、10-7mol/L)刺激肾小球系膜细胞72h后,用半定量RT-PCR法检测细胞中CTGFmRNA的表达。②以10-7mol/L的ALD刺激肾小球系膜细胞不同时间(12、24、48、72h)后,用半定量RT-PCR法检测细胞中CTGFmRNA的表达。结果半定量RT-PCR结果显示,随着ALD浓度的升高,CTGFmRNA的表达量(以CTGF/GAPDHmRNA表示)明显增加,在10-7mol/L时达高峰,与对照组比较,差异有显著性(P<0.01)。表明ALD促进肾小球系膜细胞CTGFmRNA的表达,且具有一定的剂量-反应关系。以10-7mol/L的ALD作用于大鼠肾小球系膜细胞12~72h,随着作用时间的延长,CTGFmRNA的表达量明显增加,在72h时达高峰,与对照组比较,差异有显著性(P<0.01)。表明ALD促进系膜细胞CTGFmRNA的表达,且具有一定的时间-效应关系。结论ALD促进大鼠肾小球系膜细胞CTGFmRNA的表达,从而促进慢性肾脏病进展。
[Objective] To evaluate the effect of aldosterone (ALD) on connective tissue growth factor (CTGF) mRNA expression in cultured rat mesangial cells. [Methods] (1) Mcsangial cells were cultured in medium contain- ing different concentrations of ALl) (10^-11、10^-10、10^-9、10^-8、10^-7mol/L) for 72 h, while control cells were treated with vehicle only. The expressions of CTGF mRNA were detected by semi-quantitative RT-PCR. (2) Mesangial cells were stimulated with 10-7 mol/L ALD for 12, 24, 48, 72 h. The expressions of CTGF mRNA were detected by semi- quantitative RT-PCR. [Results] Semi-quantitative RT-PCR method showed that the expressions of CTGF mRNA were increased significantly in a dose- and time-dependent manner after'stimulated by aldos'terone in cultured rat mesangial cells. [Conclusion] ALD up-regulates the expression of CTGF mRNA in cultured rat mesangial cells. ALD may play a crucial role in the progression of chronic kidney disease.
