摘要
【目的】探索鸡肠道抗菌肽的乙酸提取方法,并寻找乙酸提取的最佳条件。【方法】采用乙酸浸提法从鸡肠道中提取抗菌肽,并通过琼脂糖弥散方法检测每步产物的活性,用最小抑菌浓度(MIC)法测定提取物对巴氏杆菌的相对抑菌效价来检测提取率。【结果】从鸡肠道黏膜分离到的抗菌肽是Gal-13的前片段和成熟肽,其相对分子质量约为7.71 ku。乙酸提取的最佳条件是:乙酸4℃浸提24 h,Gal-13的提取率较高,为0.1%。【结论】鸡肠道黏膜的抗菌肽主要是Gal-13。
[Objective] The study investigated the method of purified antimicrobial peptide from chicken intestine mucosa by acetic acid extraction and optimized the extracting technology. [Method] Antimicrobial peptide was isolated from chicken intestine by acetic acid extraction. Agarose diffusion assay method was used to detect antimicrobial peptides' activity; minimal inhibitory concentration (MIC) method was employed to determine the content of antimicrobial peptides. [Result] Chicken intestine yielded of Gal-13. Gal-13 appears as propiece and mature peptide when extracted from intestine,and the molecular weight was 7.71 ku. The best extraction technology of antimicrobial peptides in chicken intestine was acetic acid extracted 24 hours at 4℃ with the extraction 0.1%. [Conclusion] Gal-13 was main antimicrobial peptide in chicken intestine mucosa.
