咳喘宁对支气管哮喘大鼠肺组织MMP-9mRNA表达的影响 被引量:4

摘要 目的观察咳喘宁对支气管哮喘大鼠肺组织MMP-9 mRNA表达的影响。方法取SD大鼠40只,随机分为正常对照组、模型对照组、咳喘宁高、低剂量组(27,13.5 g/kg)和桂龙咳喘宁组(0.41 g/kg),每组8只。除正常对照组外,其余各组以卵蛋白致敏并吸入激发法制备大鼠支气管哮喘模型,各给药组均从第1次哮喘激发开始(造模第3周)至处死前每天灌胃给药,激发并给药4周后处死大鼠,采用RT-PCR法检测MMP- 9 mRNA的表达,HE染色法观察肺和支气管病理组织学变化。结果与正常对照组相比较,模型对照组大鼠支气管管壁和平滑肌厚度、嗜酸性粒细胞、淋巴细胞数以及肺组织MMP-9 mRNA表达均明显增加(P<0.01);与模型对照组比较,各治疗组均可显著降低支气管管壁和平滑肌厚度、嗜酸性粒细胞、淋巴细胞数以及MMP-9 mRNA的表达(P<0.01)。结论咳喘宁能够降低肺组织MMP-9 mRNA的表达,抑制气道炎症和气道重塑。 Objective To observe effect of Kechuan Ning (KN) on the expression of MMP- 9mRNA in lung tissue of bronchial asthmatic rats. Methods Forty rats were randomly divided into 5 groups: the normal group, the model group, high- dose KN group (27 g/kg), low- dose KN group(13.5 g/kg), and Guilong Kechuan Ning group (0.45 g/kg), 8 rats in each group. The bronchial asthmatic model was established by ovalbumin sensitization and long -term inhalation provocation. The rats in treatment groups received gastric garage each day from the first time of provocation. After treatment for 4 weeks, the rats were killed and the pathological changes of lung tissue were observed after HE stain. The expression of MMP- 9mRNA in lung tissue was determined by RT- PCR. Result Compared with the normal group, the thickness of bronchial wall and bronchial smooth muscle, the numbers of eosinophils and lymphocytes, and expression of MMP- 9mRNA in lung tissue were increased( P 〈 0. 01); Compared with the model group, the thickness of bronchial wall and bronchial smooth muscle, the numbers of eosinophils and lymphocytes, and the expression of MMP -9mRNA in lung tissue were decreased in the treatment groups( P 〈 0.01). Conclusion Kechuan Ning could de- crease the expression to MMP - 9mRNA to restrain airway remodeling and inflammation.
