摘要
目的通过观察消瘀化痰饮对非酒精性脂肪肝(NAFLD)大鼠肝细胞凋亡及相关调控基因Bcl-2和Bax蛋白表达的影响,探讨其治疗NAFLD的作用机制。方法采用高脂饮食喂饲大鼠复制NAFLD模型,以东宝甘泰作为阳性对照药,运用流式细胞仪观察各组大鼠肝细胞凋亡及相关调控基因Bcl-2和Bax蛋白表达的变化,同时采用HE染色观察各组大鼠肝组织形态学变化。结果光镜下可见模型组大鼠肝脏中度以上脂肪变性,并有少量的肝细胞坏死;流式细胞术分析可见其细胞凋亡率明显增高(P<0.01),Bcl-2蛋白表达FI值降低(P<0.01),Bax蛋白表达FI值增高(P<0.01)。与模型组比较,各用药组肝脂变程度显著改善,肝细胞的凋亡率明显下降(P<0.01),Bcl-2蛋白表达FI值增高(P<0.01),Bax基因表达FI值降低(P<0.05或P<0.01)。结论消瘀化痰饮能使Bcl-2蛋白表达上调、Bax蛋白表达下降,抑制细胞凋亡趋势,起到抗脂肪肝的作用。
Objective To observe the effect of Xiaoyu Huatan Decoction (XHD) on hepatic apoptosis and apoptosis - related gene Bcl -2, Bax expression in nonalcoholic fatty liver disease(NAFLD)rats, and to explore its therapeutic effect for NAFLD. Methods The rat model of NAFLD was induced by feeding high fat forage. Dongbao Gantai was used as the positive control drug. Flow cytometry(FCM)was used to detect the hepatic apoptosis and expression of Bcl- 2 and Bax, and optical microscope was used to observe the degree of hepatic steatosis. Results Moderate or serious steatosis of liver cells was found under optical microscope, and there existed a few hepatic necrosis. The apoptotic rate and FI expres- sion of Bax in the model group were significantly increased ( P 〈 0. 01 ), while FI expression of Bcl - 2 was significantly decreased ( P 〈 0.01). Compared with the model group, the degree of hepatic steatosis was improved, the apoptotic rate and expression of Bax were significantly decreased, while expression of Bcl - 2 was significantly increased in the treat- ment group ( P 〈 O. 05 or P 〈 O. 01 ). Conclusion Xiaoyu Huatan Decoction has an effect on restraining fatty liver by up - regulating the expression of Bcl - 2, decreasing the expression of Bax and inhibiting hepatic apoptosis.
