摘要
目的探索以牛Ⅰ型胶原作为人工皮肤支架的可行性.方法将牛Ⅰ型胶原用醋酸溶解后,采用-70℃预冻,再采用真空干燥的方法获得作为人工真皮的胶原海绵支架.结果得到膜状的胶原海绵支架,其切面可见胶原纤维相互连接成多孔网状结构,孔径大致相同.结论牛Ⅰ型胶原是一种良好的细胞外基质,可以用来构建人工皮肤的真皮支架.
Objective To explore the possibility of using Ⅰ collagen as the scaffold to fabricate the artificial skin. Methods The Ⅰ collagen which had been solubilized into 0.5 M by acetuin was put into -70 ℃ refrigeratory overnight. The freeze-drying collagen was tested as the scaffold by microscope and scanning electron microscope (SEM) in the artificial skin. Result The scaffold was fabricated by Ⅰ collagen successfully. Conclusion The scaffold made by Ⅰ collagen is proved to be a good scaffold in the artificial skin.
