SET对大鼠生育力与早期胚胎发育毒性的研究

摘要 目的评价SET对SD大鼠生育力及早期胚胎发育毒性的作用.方法将160只SD大鼠(雌雄各半)随机分为溶媒对照组和SET0.125、0.25、0.5g/kg的3个剂量组.雄、雌鼠分别连续给药8周,2周后按1:1合笼,共交配5d,交配期间每日清晨进行阴道涂片检查有无精子,查到精子当天记为妊娠第0d;雄鼠给药至交配成功后处死,雌鼠给药至妊娠第6d,于妊娠第15d处死,进行生育力及早期胚胎发育毒性相关指标的检测.结果试验期间各组动物体重增长,进食量正常;动物交配、生育能力及早期胚胎发育未见明显异常,组间比较无统计学意义(P>0.05).结论批号为20051101的SET,在本试验剂量范围(0.125、0.25、0.5g/kg)对SD大鼠生育力及早期胚胎发育未见明显毒性作用. Objective To evaluate the fertility and early embryonic development toxicity on SD rats. Methods We divided 160 SD rats randomly into solvent control and three dose groups that are 0.125, 0.25 and 0.5 g/kg. Male rats are administrated by gavages for 8 weeks continuously and female rats for two weeks, then delivery them into the same cage by 1:1 to mates for five days. During this period, in every morning we did the vaginal smears test to check in the presence of sperm. The sperm was found and took down the day of gestation day 0. The male rats mate successfully were killed. The female rats were administrated to the sixth days of pregnancy and were killed in the 15th days of pregnancy. Then we anatomized, observed and tested the indicators. Results Body weights of rats grow, food intake was normal. The animals' fertility was no obviously abnormal. Animal testicles, epididymis, the uterus, ovary and weight coefficient between the two groups were no significant differences (P 〉 0.05). Early embryonic development was no obvious abnormalities. Conclusion Rats' fertility and early embryonic development is no obvious toxicity in the test dose range of 20051101 batches of SET, that is 0.125, 0.25 and 0.5 g/kg by body weight.
