高效微生物絮凝剂产生菌DS16发酵最优化条件研究

摘要 筛选到一株产微生物絮凝剂的细菌DS16,研究发现其最佳发酵时间为18h。通过正交试验对发酵生产进行最优化研究,确定其最佳培养基成分为K2HPO40.45%、NaH2PO40.14%、液糖1.8%、(NH4)2SO40.5%、酵母提取物0.04%、Mg2SO40.02%、CaCl20.01%;最佳培养条件为温度29℃、摇瓶转速为170rpm、通气量为0.2V/V·min、投料系数为0.76。Mn2+对絮凝剂的产生有显著的促进作用,在添加量为6mg/L时絮凝剂产量达到最高,获得最高絮凝活性的使用量仅为2mg/L。 A microbial flocculant--producing strain (DS 16) was screened. The optimum fermentation period of DS 16 to produce flocculant was 18h. Fermentation optimization was studied by orthogonal experiment ,and the optimun component proportions of medium obtained were: K2HPO4 0.45%, NaH2PO4 0.14%, liquid sugar 1.8%, (NH4) 2SO4 0.5%, yeast extract 0.04%, Mg2SO4 0.02%, CaCl20.01%: the optimum culture conditions were: temperature 30℃, agitation rate 170 rpm, air volume 0.2VN-min, loading coefficient 0.760 Mn2 + can promote notablely the microbial flocculant production. The highest output is achieved when the adding content of Mn2+is 6mg/L, and the used dosage for getting maximal flocculating activity is only 2mg/L Mn2+ .
