摘要
目的 观察伏格特-小柳-原田(VKH)综合征患者治疗前以及治疗1个月后外周血中CD4^+CD25^+FOXP3^+T细胞的比例变化。方法采集15例VKH综合征患者治疗前以及系统的糖皮质激素治疗1个月后的外周血,分离淋巴细胞。采用细胞表面标志物CD4、CD25以及特征性的细胞核内转录因子叉状头/翅膀状螺旋转录网子(FOXP3)的抗体标记CD4^+CD25^+调节性T细胞,流式细胞仪检测CD4^+CD25^+FOXP3^+T细胞的比例,并与15例以健康志愿者为正常对照组的外周血中的该亚群细胞水平进行比较。结果外周血中CD4^+CD25^+FOXP3^+T细胞占CD4^+细胞的比例,治疗前患者组为(0.30±0.19)%,正常对照组为(1.41±0.52)%,二者比较,差异有统计学意义(t=7.665,P〈0.01);治疗1个月后,患者组为(1.28±0.54)%,接近正常对照组。另有2例患者在治疗1个月后外周血中CD4^+CD25^+T细胞比例异常升高。结论VKH综合征患者外周血中CD4^+CD25^+FOXP3^+T细胞的比例治疗前显著低于正常对照组。治疗后与正常对照组接近,表明VKH综合征的发病可能与患者外周血中天然性CD4^+CD25^+调节性T细胞的比例下降有关。
Objective To observe the peripheral blood of patients with Vogt Koyanagi proportion changes of CD4^+ CD25^+ FOXP3^+ T cells in -Harada disease (VKH) before and after one month of treatment. Methods The peripheral blood samples from 15 patients with VKH disease before and after one month of treatment by glucocortieoid, and from 15 healthy volunteers were collected ,and lymphocytes were separated from them. CD4^+ CD25^+ regulatory T cells were labeled by antibodies of cell surface marker CD4,CD25 and transcription factor FOXP3. The proportion of CD4^+CD25^+FOXP3^+ T cells were detected by flow cytometry. Results Before the treatment, the percentage of CD4^+CD25^+FOXP3^+ T cells in periphery blood was (0.30±0.19)% of CD4^+ ceil in VKH patients, and (1.41 ±0.52)% in control group, the difference was statistically significant (t= 7. 665,P〈0.01) ; after one month of treatment, the VKH patients group was (1. 28±0. 54 ) % which close to the control group. However there were two patients whose CD4^+CD25^+ T cells increased extraordinarily after one month of treatment. Conclusions The proportion of CD4^+CD25^+ FOCP3^+ T cells in periphery blood in VKH patients were lower than control group obviously before treatment, but were close to control group after treatment. Those results indicated that VKH diseases may be associated with the decreased proportion of CD4^+CD25^+ regulatory T cells.
