摘要
目的:研究RASSF1基因在各种肿瘤细胞中的表达情况以及分析RASSF1各成员与肿瘤发生的相关性。方法:设计特异性的引物,用RT-PCR方法检测5种肿瘤细胞中RASSF1家族7种不同转录本mRNA的表达情况。结果:同源序列分析表明,转录本A、D、E序列最长,同源性较高,推测是起主要功能的一类基因,而B、C、F和G等缺失部分较大,可能是细胞突变引起;转录本A、D和E在各种肿瘤细胞中均未见表达,而其他转录本B、C和F在肿瘤细胞中有转录。结论:RASSF1基因7种转录本在被测试的5种肿瘤细胞中的表达存在明显差异,转录本RASSF1的A、D和E等在几种肿瘤细胞中均未表达,说明这几个转录本可能作为抑癌的候选基因。
Objective To study the expression of RASSF1 gene in several cell lines and analyze the relationship between transcripts of RASSF1 gene and carcinogenesis. Method The expression of RASSFI gene in five tumor cell lines was detected by RT-PCR with anchored primers. Results Homology analysis showed that transcript A, D and E had the longest sequence and high homology, indicating that they were the genes that played major roles, transcript B, C, F and G had many sequences deleted, which may result from mutations in the cell. No transcript A, D, E was detected in the tumor cell lines examined, however, transcript B, C, F were detected. Conclusion There is significant difference in the expression of seven transcripts of RASSF1 gene in the five tumor cell lines examined. No transcript A, D, E is detected in the tumor cell lines examined, suggesting that these transcripts may be candidate genes for inhibiting cancers.
