共刺激分子B7-H3在肠癌中的表达及其临床意义

摘要 目的探讨肠癌组织中共刺激分子B7-H3的mRNA表达水平与肠癌发生、发展及淋巴结转移的关系。方法Real—time RT—PCR检测22例肠癌及其临近正常组织中B7-H3基因和内参照GAPDH基因的mRNA表达水平,并求出两者比值,记为B7-H3N,同时计算同一患者肠癌组织与其正常组织中B7-H3N的比值,定义为B7-H3T/N,分析B7-H3T/N与肠癌患者临床参数的相关性。结果肠癌组织中B7-H3T/N的表达水平与淋巴结转移及NM23有显著相关性。结论通过检测肠癌组织中B7-H3的mRNA表达水平可以评价肠癌的淋巴结转移及预后。 Objective We detected the mRNA expression level of B7 - H3 in colorectal carcinoma tissues and adjacent normal tissues, and examined the correlation between B7 - H3 mRNA expression and oncogenesis, development and metastasis of colorectal carcinoma. Methods Real- time RT- PCR was applied to analyze B7- H3 and GAPDH mRNA expression level in 22 coloreetal carcinoma tissues and corresponding adjacent normal tissues. B7 - H3 mRNA expression difference in carcinoma tissues and adjacent normal tissues were analyzed.The ratio of B7 - H3 to GAPDH mRNA expression level were calculated and marked as B7 - H3N, the ratio of B7 - H3N in tumor tissues to corresponding normal tissues were subeequendy calculated and marked as B7 - H3T/N, and the correlation between B7 - H3T/N and patients' clinical parameters was analyzed. Results The B7 - H1T/N was significandy related with patients' lymph node metastasis and NM23. Conclusion The detection of B7 - H3 mRNA expression level of in colon carcinoma tissues was beneficial for us to evaluate the lymph node metastasis and prognosis of colorectal carcinoma.
