摘要
目的:探讨中药夜明方对遗传性视网膜色素变性小鼠的治疗作用。方法:选4周龄和12周龄rds小鼠各9只,各随机分为3组,即空白对照组、杞菊地黄丸组和中药夜明方组,每组3只小鼠。夜明方组每天20 mL/kg夜明颗粒溶液灌胃,杞菊地黄丸组每天20 mL/kg杞菊地黄丸原液灌胃,空白对照组每天等量蒸馏水灌胃。连续28天灌胃后,颈椎脱臼术处死小鼠,摘取眼球,立即经10%甲醛固定48 h,光学显微镜下观察水平视网膜近赤道部位光感受器细胞的厚度。结果:观察结果显示,经夜明方治疗28天后,与空白组和杞菊地黄丸组相比较,夜明方可延缓rds小鼠视网膜外核层层数的减少(P<0.05)。结论:夜明方可延缓rds小鼠视网膜光感受器的破坏。
Objective: To approach the curative effect of Formulae of Sensitive Night Vision on heritage pigmentosa retinae through observing the pathological changes of photoreceptor ceils of rds mouse' s amphiblestrodes. Methods: 9 rds mice of 4 weeks' old and 9 rds mice of 12 weeks' old were divided into three groups randomly, which were blank control group, Lycii and Chrysanthemi and Rehmanniae Bolus group, and Formulae of Sensitive Night Vision group, three mice each group. In Formulae of Sensitive Night Vision group, oral liquor of Granule Sensitive Night Vision was given to the mice as 20 mL/kg each day by intragastric administration. In Lycii and Chrysanthemi and Rehmanniae Bolus group, stock solution of Lycii and Chrysanthemi and Rehmanniae Bolus was given to the mice as 20 mL/kg each day by intragastric administration. In blank control group, Distilled Water was given to the mice as the same dose by intragastric administration. After continuous 4 weeks' intragastric administration the mice were put to death by luxation, and their eye globes were selected. Immediately eye globes were dealt with 10% formalin fixation. To observe the thickness of photoreceptor cells near equatorial locus in level amphiblestrodes under light microscope. Results: Pathological result displayed that Formulae of Sensitive Night Vision could delay the layer numbers of outer nuclear layer of retina of rds mice after 28 days' treating of Formulae of Sensitive Night Vision,contrasting to blank control group and Lycii and Chrysanthemi and Rehmanniae Bolus group (P 〈 0. 05 ). Conclusions : Formulae of Sensitive Night Vision can delay the demolition of amphiblestrodes photoceptor of rds mouse.
