摘要
目的研究顺铂(cDDP)作用下人卵巢上皮性癌细胞株3AO中肿瘤转移抑制基因KAI1/CD82的表达的变化,初步探讨其作用机制。方法采用倒置显微镜观察cDDP作用后3AO细胞形态学的变化;四甲基偶氮唑蓝(MTT)法观察cDDP对细胞增殖的影响;运用免疫细胞化学方法检测不同浓度cDDP(1、3、9 mg/L3个浓度组)作用的3AO细胞株中KAI1/CD82表达情况,并与细胞对照组(未经药物处理)进行比较。结果cDDP作用后细胞体积缩小、细胞内颗粒增多,细胞核固缩、深染,细胞凋亡;cDDP对3AO细胞的生长抑制作用呈明显的剂量依赖关系;cDDP作用后卵巢癌细胞中KAI1/CD82基因的表达较对照组明显增强。结论cDDP不仅能增加对卵巢癌细胞的生长抑制率,而且能上调卵巢癌细胞中肿瘤转移抑制基因KAI1/CD82的表达,为临床上药物抑制肿瘤转移的深入研究提供了有利的实验依据。
Objective To investigate the effect of cisplatin (cDDP) on variations of the expression of metastasis suppressor gene KAI1/CD82 in human ovarian epithelial cancer cell line 3AO and to make a preliminary study of the mechanism of cDDP. Methods Following cDDP administration, the 3AO cells was visualized morphologically by inverted microscopy. Methyl thiazolyl tetrazolium (MTr) assay was employed to observe the effects on cellular proliferation. A wide range of eDDP concentrations ( 1 mg/L, 3mg/L and 9 mg/L) were used to detect the expression levels of KAI1 gene in 3AO cell lines by immunocytochemistry (ICH) and to make comparisons with the control group (untreated with the drug). Results Treated with all concentrations of cDDP, 3AO cells shrinked, cellular granule number increased, resulting in nuclear pycnosis, deep - staining and cell apoptosis. MTT assay demonstrated that cDDP could partially inhibit the growth of 3AO cells, with a marked dose - dependent tendency. The KAI1/CD82 protein expression in different 3AO cells treated with cDDP was significantly up - regulated compared with the control ( P 〈 0.05 ). Conclusion Not only does cDDP have a significant inhibitory effect on the growth of human ovarian cancer cell line, but also could effectively up - regulate KAI1/CD82 expression in human ovarian cancer cell line, which provides beneficial experimental evidence for clinical chemotherapy for tumor metastasis.
