食品中沙门菌变性高效液相色谱检测技术的研究与方法建立 被引量:4

摘要 目的:应用PCR结合变性高效液相色谱(DHPLC)技术建立食品中沙门菌的快速检测方法。方法:根据沙门菌fimY基因序列的特点设计特异性引物,PCR扩增的产物经DHPLC技术进行快速检测。以沙门菌等89株参考菌株做特异性检测;沙门菌液体培养物稀释成不同梯度,做灵敏度检测。结果:试验结果表明该方法有很好的特异性,而其它非沙门菌均为阴性。该方法灵敏度较高,检测低限可达到45 cfu/ml。结论:该方法可以快速、准确检测食品中沙门菌,是食品中致病菌检测的新技术和新方法。 Objective:To identify the fimY gene in Salmonella,a DHPLC assay was performed in this study.Methods:Primers specific for the fimY gene in Salmonella of food were selected to conduct the DHPLC assays.The specific testing was performed with Salmonella and 89 strains,and various grads of Salmonella was used for the sensitivity testing.Results:The good specificity was found in the study,while other non-Salmonella were negative.Meanwhile,the method showed nicer sensitivity with the lowest amount of detecting was 45 cfu/ml,so it can detect and identify Salmonella quickly and correctly.Conclusion:The DHPLC could be a new method for detecting food pathogens.
