摘要 目的:建立血清样品中毒鼠强的气相色谱/质谱联用(GC-MS)检测方法。方法:血清样品经乙酸乙酯提取、离心、浓缩、定容后,采用EI源在选择离子模式下进行检测。对提取溶剂类型、用量及质谱分析参数等因素进行了考察。结果:确定用乙酸乙酯作为提取剂,用量为6 ml,提取时间为30 min,提取次数为2次。毒鼠强工作曲线的线性范围为0.5-20μg/ml,相关系数r=0.9991,空白血清加标平均回收为80.7%,测定结果相对标准偏差为2.81%。结论:该方法回收率高、重现性好、灵敏度佳、干扰少,满足中毒样品中的毒鼠强的定性定量检验要求。 Objective:To develop a method to extract and determine tetramine in serum.Methods:After extracted and concentrated,serum sample has been monitored by GC-MS.Some factors effecting extracting tetramine were considered,including solvent type,solvent dosage and parameters of GC-MS.Results:Ethyl acetate was determined to be the solvent dosage.The extraction was carried out for two times for 30 min with 6 ml dosage.The tetramine response was linear in the range of 0.5-20 μg/ml,the correlation coefficient was 0.9991,the average recovery in serum was 80.7%,RSD was 2.81%.Conclusion:The proposed method had high recovery,good reproducibility,good sensibility,little disturbance and has been proved to be satisfactory for qualitative and quantitative need that is required in poisoning sample.
