摘要
为探讨Leptin对小鼠皮肤创伤的保护作用机制,本文采用昆明小鼠40只,在背部各制备4个全层皮肤缺损创面(0.8cm×0.8cm),将创面分成4组,分别用mLeptin、mLeptin-Ab、mLeptin+mLeptin-Ab、生理盐水局部涂抹,测定伤口愈合时间和愈合率,并通过RT-PCR对伤口组织Leptin、Leptin受体(OB-Rb)和EGF-R的表达进行了检测。结果表明:与生理盐水对照组比较,mLeptin组和/或mLeptin-Ab组小鼠皮肤伤口愈合时间明显缩短(P<0.01),EGF-R的表达明显升高(P<0.01);单用mLeptin-Ab组小鼠皮肤伤口愈合率,EGF-R、OB-Rb和Leptin的表达均显著提高(P<0.01)。结论为Leptin和/或mLeptin-Ab均可促进小鼠皮肤伤口愈合;未发现mLeptin-Ab对Leptin的中和作用,单用mLeptin-Ab反而能够促进伤口修复。
To assess the possible protective effect of Leptin and its antibody against mice impaired wound,forty Kunming mice,who received full thickness skin wound(0.8 cm×0.8 cm) in each side of midline were obtained.The wound mice were divided into four groups and treated with mLeptin,mLeptin-Ab,mLeptin plus mLeptin-Ab and physiological saline respectively.The healing time and the healing rate of wound in mice were measured.The expression of Leptin,Leptin receptor(OB-Rb) and EGF receptor in wound tissue were observed by RT-PCR.Compared with control group,the healing time in Leptin and/or Leptin-Ab treated groups was significantly shortened(P〈0.01),the levels of EGF receptor expression were increased markedly(P〈0.01).In Leptin-Ab treated group,the healing rate was predominantly elevated(P〈0.01),and the expression levels of EGF receptor,OB-Rb and Leptin were increased significantly.Therefore,the neutralization of Leptin-Ab against Leptin was not found in this study,however,it suggested that the topical application of Leptin and Leptin-Ab could significantly accelerate the healing of impaired wound in mice,and the possible protective effect of Leptin-Ab may be higher than that of Leptin.
