^(125)Ⅰ标记的Flt4多抗导向定位淋巴结的可行性分析及其代谢动力学研究

摘要 探讨125I标记的Flt4多抗(125I-Flt4PcAb)导向定位淋巴结的可行性及其在体内的代谢动力学特点,为应用Flt4PcAb进行前哨淋巴结(sentinel lymph node,SLN)的检测提供实验依据。应用氯胺T法对Flt4PcAb进行125I标记;将125I-Flt4PcAb经大鼠足背皮下注射,研究125I-Flt4PcAb在淋巴结内浓集及其在体内的代谢动力学特点。结果表明125I-Flt4PcAb经皮下注射后,在腘窝淋巴结的ID(%)/mg值最高,血液次之,其它各组织器官均很小;腘窝淋巴结的ID(%)/mg值在注射后0.5h达最高,血液32h达最高;其在体内的代谢可进行二室动力学模型拟合,吸收相半衰期(T1/2Ka)为0.6±0.1d,清除相半衰期(T1/2β)为3.5±0.5d。这提示125I-Flt4PcAb能够对淋巴结进行导向定位;它在体内的代谢符合二室动力学模型,清除相半衰期较长。 To analyse lymph node guide-localization ability the metabolic kinetics of ^125I-Flt4 PcAb in order to provide experimental basis for localization of SLN with Flt4 PcAb,Flt4 PcAb was labeled by ^125I via chloramine T method.^125I-Flt4 PcAb was administered via subcutaneous injection at dorsum of foot in rats in order to test whether ^125I-Flt4 PcAb would gather in lymph node and provide the metabolic kinetic trait of ^125I-Flt4 PcAb in vivo.The results showed that the ID(%)/mg of popliteal lymph node was highest and blood higher and the other organs low.The ID(%)/mg of popliteal lymph node was highest at 30min after ^125I-Flt4 PcAb injection and blood 32h.^125I-Flt4 PcAb metabolism after subcutaneous injection met a two compartment fitted model.The mean value of T l/2Ka and T1/2β was 0.6±0.1d and 3.5±0.5d respectively.It suggested that ^125I-Flt4 PcAb was a potential agent for guide-localization of lymph node.Its metabolism met a two compartment model and T1/2β was long.
