摘要
水稻(Oryza sativa L)耐淹品种FR13A在分蘖期进行没顶淹涝处理。分别取淹涝0h,2h,4h,8h,16h,32h的叶片进行mRNA差异显示分析,获得8个差异表达的cDNA片段。利用反式Northern斑点杂交法去除4个假阳性片段,对4个阳性片段(DF8、DF9、DF10、DF11)测序后进行同源性分析,其中一个片段(DF8)的蛋白产物与WD-40重复蛋白高度同源,另一个片段(DF11)的蛋白产物与植物生长素应答蛋白有较高同源性,其余2个没有找到同源蛋白质序列。
The submergence-tolerant rice variety FR13A (Oryza sativa L) was treated with complete submergence at tillering stage. The leaves which were completely submerged for 0h,2h,4h,8h,16h and 32h,were sampled to execute mRNA display respectively. Eight cDNA fragments were isolated by mRNA differentially display reverse transcription PCR (DDRT-PCR),but only four fragments were approved truth by reverse Northern blotting. These fragments named as DF8,DF9,DF10 and DF11 were involved in sequencing and analyzed by Blast program. The result showed that the putative protein of DF8 was highly homologous with WD-40 repeat protein and that of DF11 was homologous with auxin responsive GH3-like protein. Two rests did not found homologous proteins.
