摘要
建立了油菜籽中VE的高效液相色谱测定方法,比较了样品处理方法对测定油菜籽中VE的影响。采用硅胶柱色谱柱,流动相为正己烷∶乙醚=93∶7(v/v),流速为1.2 mL/m in,荧光检测器激发波为290 nm、发射波长为330 nm。线性范围为10-60μg/mL,平均回收率为107%,变异系数为1.1%,正己烷溶剂浸提法比皂化方法好。
A method of high pressure liquid chromatography(HPLC) for determining VE in rapeseeds was developed, and the effect of different sample preparation on the determination was discussed. The determination conditions of the developed method are as follows : SUPELCOSILTM LC - SI column, mobile phase hexane : ether = 93 : 7 (v/v), flow rate 1.2 mL/min, and a fluorescence detector at λ ex = 290 nm and λ em = 330 rim. The linear range of calibration curves is 10 - 60 μg/mL, the average recovery rate is 107%, and the relative standard deviation is 1.1% . The procedure of hexane extraction for sample preparation is better than the saponification method.
