摘要 通过对传统的双向电泳技术进行改进和优化,建立适合分析黄花苜蓿蛋白组学研究的双向电泳体系。结果表明,使用改进方法后所获得的双向电泳图谱稳定性和重复性较好,分辨率较高,经银染后可分辨出400多个蛋白点,其等电点主要分布于pI5~7,分子量主要分布于10~150kDa;经考马斯亮蓝染色后可分辨出800多个蛋白点,其等电点主要分布于pI5~7,分子量主要分布于10~150kDa。 Based on the conventional method of O'Farrell, an improved procedure for the two--dimensional gel electrophoresis of Medicago falcata L. was presented. The two--dimensional separation of proteins was distinct and steady. The detected protein spots stained by silver nitrate were more than 400, with iso- electric points(pI) ranging mainly from 5 to 7 and molecular weights(MW) from 10 kDa to 150 kDa. Using Coomassie brilliant blue G-250, the detected protein spots stained were more than 800, with isoelectric points(pI) ranging mainly from 5 to 7 and molecular weights(MW) from 10 kDa to 150 kDa.
