大黄对百草枯中毒大鼠急性肺损伤保护作用的研究 被引量:10

摘要 目的:观察大黄对百草枯(PQ)中毒大鼠急性肺损伤的保护作用,探讨其可能的作用机制。方法:168只SD大鼠随机分为对照组、染毒组、大黄干预组和大黄干预对照组。PQ(50 mg/kg)灌胃染毒,生大黄(300 mg/kg.d-1)干预。观察肺组织病理变化,测定肺泡灌洗液中细胞计数、蛋白含量和血清超氧化物歧化酶(SOD)、丙二醛(MDA)含量,计算肺泡灌洗液中中性粒细胞百分比及肺泡通透指数。结果:①染毒组组织充血、水肿,炎性细胞浸润,少数见纤维化改变,肺泡通透性增加,而大黄干预组上述改变减轻。②血清SOD活性:染毒6 h后染毒组降低,与对照组比较有显著性差异(P<0.05),大黄干预组升高,48 h达峰值并维持在较高水平,与染毒组比较有显著性差异(P<0.05)。大黄干预对照组升高,但升幅低于大黄干预组,两组比较有显著性差异(P<0.05)。③血清MDA活性:染毒组、大黄干预组均升高,大黄干预组升幅小于染毒组,但无统计学意义。结论:大黄对PQ中毒大鼠急性肺损伤具有保护作用,其机制可能是减轻了肺组织的脂质过氧化反应。 Objective: To evaluate the potential alleviating effect of rhubarb on acute lung injury caused by paraquat(PQ) in rats and its mechanism. Methods:A total of 168 healthy adult SD rats were randomly divided into Group A ( blank control), B ( poisoned ), C ( rhubarb-treated ) and D ( rhubarb-treated control). Group B and C were treated intragastrically with PQ at 50 mg/kg ,while C and Group D given rhubard at 300mg/(kg · d), followed by observation of the pathological changes of the lung tissues and determined the count of neutrophils and the contents of proteins, serum superoxide dismutase (SOD) and malondialdehyde (MDA) in the pulmonary alveolar lavage fluid. Results : Congestion, edema, leukocyte infiltration and increased pulmonary alveolar permeability were observed in Group B, with fibrosis in a few rats. All the changes mentioned above were obviously alleviated in Group C . The level of serum SOD at 6 hours after exposed to PQ were markedly reduced in Group B as compared with Group A (P 〈 0.05) ;but it was increased in Group C, reached the peak value and maintained a high level after 48 hours ,with a statistical difference from Group B, (P 〈0.05). So was it in Group D , though h'ss obviously than in Group C, with a significant difference in between ( P 〈 0.05 ). A rise in the level of serum MDA was noted in Group B and C, by a bigger margin in the former,but with no significant difference. Conclusion: Rhubarb alleviates acute lung injury in rats with PQ poisoning,which may be attributed to its attenuation of lipid peroxidation in the lung tissues.
