皮肌炎患者外周血CD4^+Th1/Th2细胞因子及转录因子的表达 被引量:1

摘要 目的探讨Thl/Th2细胞分化趋势在活动性皮肌炎(DM)发病机制中的作用。方法分别采集15例活动性DM患者(DM组)和15例健康人(对照组)外周血,采用免疫磁珠分离获得CD4+T细胞(Th细胞),逆转录聚合酶链反应法(RT-PCR)检测两组Thl、Th2转录因子T-bet、GATA3及细胞因子γ干扰素(IFN-γ)、白细胞介素4(IL-4)的表达强度。分析T-bet和IFN-γ、GATA3和IL-4的相关性;IFN-γ、IL-4表达与血清肌酸激酶(CK)含量的相关性。结果DM组IFN-γ、GATA3及IL-4表达均明显高于对照组(P<0·05);DM组IL-4与GATA3、血清CK呈正相关(P<0·05)。结论DM患者Thl/Th2无明显极化现象,Thl/Th2细胞因子表达均增强。 Objective To investigate differentiation of Th1 /Th2 in pagients with dermatomyositis (DM). Methods CD4^+ T cells were isolated from peripheral blood of 15 DM patients (group A) and the 15 controls (group C) by microbeads. The gene expressions of Th1/Th2 type cytokines of interferon fector-Y(IFN-Y) and interleukin-4(IL-4), transcription factors T-bet and GATA3 were determined by RT PCR. Results The expression levels of IFN-7,GATA3 and IL-4 in DM patients were higher than those in group C (P〈0.05). There was a positive correlation between IL-4 and GATA3, IL-4 and serum CK in DM patients (P〈0.05). Conclusion The expression of Th1 /Th2 type cytokines is increased and there is no obvious polarization in Th1/Th2 in the patients with DM.
