白介素-1β上调肺腺癌细胞系A549 VEGF的表达

摘要 目的探讨IL-1β调节肺腺癌细胞A549血管内皮生长因子(VEGF)表达的机制。方法用不同浓度的IL-1β作用于A549细胞不同时间,再用不同浓度的塞来昔布干预IL-1β诱导的A549细胞,用反转录-聚合酶链反应(RT-PCR)测定VEGF mRNA表达,用ELISA法测定培养上清液中VEGF蛋白质表达。结果IL-1β干预A549细胞后VEGFmRNA和蛋白质表达均较对照组显著增加,呈剂量依赖性(P<0.05),24h时达高峰。不同浓度的塞来昔布显著缓解上述变化(P<0.05)。结论缓解IL-1β诱导的A549细胞VEGF表达,其调节机制可能发生在转录水平。 Objective To investigate the regulation effect of interleukin-1β(IL-1β) on the expression of VEGF in A549 eells. Methods A549 cells were incubated with IL-1β in the presence or absence of Celeeoxib. After incubation, cells were harvested to detect VEGF mRNA by reverse transeription-polymerase chain reaction (RT-PCR), and the supernatant was collected to determine the concentration of VEGF protein by enzyme-linked immunosorbent assay (ELISA). Results After treatment with IL-1β, the expression of VEGF mRNA and protein in A549 cells were signifieantly higher than that in the eontrol group ( P 〈 0. 05 ) ; IL-1β up-regulated VEGF mRNA and protein expression in a dose-dependent and time-dependent manner in A549 eells, after ineubation for 24 hours, VEGF expression was inereased. After Celecoxib intervention, VEGF expression deereased as eompared with that of IL-1β induced group, the differenee was statistically significant ( P 〈 0. 05 ) ; with the concentration of Celeeoxib stepping up, VEGF expression was decreased in dose-dependent manner and the difference was signifieant( P 〈 0. 05 ). Conclusion IL-1β upregulates VEGF expression in A549 cells, the meehanism may be at transcriptional level; Celecoxib down-regulates IL-1β induees VEGF expression in A549 cells.
