摘要
对比研究高脂血症动物模型中,摄入高剂量不同富硒植物对大鼠硒累积和过氧化脂质等肝指标的影响,为指导科学安全利用植物硒预防和治疗高脂血症提供基本实验依据。采用高脂饲料诱发大鼠高脂血症模型,设计富硒大蒜和富硒绿羽治疗组和预防组,分别以高剂量两种富硒植物悬浊液连续灌胃1个月和3个月,测定和对比分析大鼠肝脏硒累积、丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GPx)活性。结果发现长期高脂饲料喂养显著降低大鼠肝脏硒浓度,摄入富硒绿羽的大鼠肝硒累积显著高于摄入富硒大蒜的,预防组与治疗组大鼠肝组织硒累积没有显著差异,证明大鼠肝脏组织硒累积主要取决于摄取的富硒植物硒种类,高剂量两种实验用富硒植物硒对高脂血症模型动物肝脏不存在安全危害;与高脂饲料模型对照组相比,补充植物硒能够在一定程度上降低大鼠肝MDA水平,显著提高肝GPx活性,表明植物硒可通过GPx清除肝组织MDA,防止长期高脂饲料引起的肝脏氧化损伤;富硒大蒜预防组和治疗组大鼠肝MDA含量存在显著差异,但两组大鼠肝GPx活性差异不显著,推断两种富硒植物硒清除肝脏MDA的机制不完全一致。
The hyperlipoidemia rat model induced by high-fat diet was applied to compare the inference of selenium(Se)-enriched garlic and kale on Se accumulation and lipid peroxidation in rat liver. Sixty adult male Wistar rats were fed the basal diet for a week, divided equally into the control, high-fat diet group ( HFD), FD + Se-enriched garlic preventive group, HFD + Se-enriched green kale preventive group, HFD + Se-enriched garlic therapeutic group and HFD + Se-enriched green kale therapeutic group. Rats were given 300 ug Se/kg bw of daily suspension of Se-enriched plants by ,gavage for 3 months in two preventive groups, and for the 3rd month in two therapeutic groups. Rats were sacrificed at the end of 3rd month, the Se accumulation, the maleic dialdehyde (MDA) contents, and the activities of glutathione peroxidase (GPX) in liver were investigated. The results showed that liver Se contents significantly decreased in rats of the HFD group, and the liver Se accumulation in rats administering Se-enriched green kale were significantly higher than those in rats administering Se-enriehed garlic, and no significant differences of liver Se accumulation were found in rats between preventive and therapeutic groups, indicating that the selenium accumulation in rat livers are more closely related to plant Se components and that high doses of two plants are safe to rat liver; The liver GPx activities in rats administering selenium-enriched plants were significantly higher than those in HFD group, but no significant decreases of liver MDA level occurred in rats of two preventive groups; The liver peroxidation could be protected by GPx, and two plants might act in different mechanisms in the elimination of the liver MDA.
