胃癌中iNOS与HIF-1α表达的相互关系及临床意义 被引量:2

Relationship between hypoxia-inducible factor-1α and inducible nitric oxide synthase expression in gastric cancer and its clinical significance

摘要 目的:研究诱导型一氧化氮合酶(iNOS)与缺氧诱导因子-1α(HIF-1α)在胃癌中表达的相互关系,探讨他们在胃癌的发生发展、浸润和转移中的作用.方法:2001-01/2002-03手术切除,病理证实为腺癌的胃癌标本54例,患者术前均未行化疗或放疗,另取正常胃组织15例作对照.应用免疫组化SP法检测胃癌及正常胃组织中iNOS和HIF-1α的表达,光镜下观察iNOS及HIF-1α蛋白表达,分析iNOS与HIF-1α的表达关系及临床意义.结果:胃癌中iNOS和HIF-1α的表达明显高于正常胃组织(66.67%vs6.67%P<0.05;74.07%vs0%P<0.01),其表达与胃癌TNM分期、肿瘤浸润深度和淋巴结转移显著相关(P<0.05或0.01).胃癌组织中iNOS与HIF-1α的表达呈正相关(r=0.596,P<0.05).结论:iNOS和HIF-1α在胃癌的发生发展中起重要作用,联合检测可作为判断胃癌恶性程度和预后的指标. AIM: To investigate the expression of inducible nitric oxide synthase (iNOS) and hypoxia-inducible factor-1α (HIF-1α) as well as their correlation in gastric cancer, and to explore their roles in the genesis, invasion and metastasis of gastric cancer. METHODS: Immunohistochemical technique was used to detect the expression of iNOS and HIF-1α in 54 cases of gastric cancer and 15 cases of normal gastric tissues. The relationships between the expression of iNOS, HIF-1α and tumor angiogenesis, clinical pathologic features of patients were analyzed. RESULTS: The expression levels of iNOS and HIF-1α in gastric cancer were significantly higher than those in the normal gastric tissues (66.67% vs 6.67%, P 〈 0.05; 74.07% vs 0%, P 〈 0.01); iNOS and HIF-1α expression in gastric cancer were significantly associated with TNM staging, invasive depth and lymph node metastasis (P 〈 0.05 or 0.01). There was a positive correlation between iNOS and HIF-1α expression (r = 0. 596, P 〈 0.05). CONCLUSION: Inducible NOS and HIF-1α play very important roles in the genesis and development of gastric cancer, and they may serve as markers in the diagnosis of gastric cancer and estimation of prognosis.
