利用蛋白转导技术将α-烯醇化酶导入心肌细胞的实验研究

摘要 目的探讨将外源α-烯醇化酶导入心肌细胞的可行性,为后续的蛋白功能研究奠定基础。方法原代培养WKY乳鼠心肌细胞,将纯化的α-烯醇化酶与蛋白转导载体Charoit共孵育后转染心肌细胞,同时设置阳性对照β-半乳糖苷酶转染组,光镜下计数转染阳性细胞的百分率;蛋白免疫印迹检测细胞总蛋白中α-烯醇化酶的表达;荧光共聚焦显微镜下观察α-烯醇化酶的亚细胞定位。结果Charoit能够将外源蛋白成功导入原代培养的心肌细胞中,光镜下可见β-半乳糖苷酶的导入效率约为81%,Western blot结果表明外源α-烯醇化酶成功导入了心肌细胞,免疫荧光显示导入的外源α-烯醇化酶与内源α-烯醇化酶相似,主要定位于胞浆。结论利用Charoit介导的蛋白转导技术能够成功将外源α-烯醇化酶导入心肌细胞。
