高效液相色谱法测定兔血浆中黄芩苷的含量

摘要 目的:建立测定兔血浆中黄芩苷的含量测定方法。方法:用PEG400—2%偏磷酸沉淀血浆蛋白,离心后取上清液进行色谱分析。色谱柱为Diamosil—C18,流动相为乙腈-0.1%磷酸(25:75,V/V),流速为1ml/min,检测波长275nm。结果:黄芩苷的质量浓度在0.08722~2.9075mg/L(r=0.9899);低、中、高3个浓度的质量控制(QC)样品的相对回收率分别为96.3%、97.3%和96.6%。结论:方法简便快速,适用于黄芩苷的血药浓度测定及药代动力学研究。 Objective: To establish a biopharmaceutical analytical method for the determination of baicalin in plasma by RP-HPLC. Methods: Rabbit plasma proteins were precipitated with PEG400- 2% metaphosphoric acid and the supernatant liquid obtained from the serum by centrifuge was subjected to chromatographic analysis. The analytical column was Eclipse XDB-C18, and the mobile phase consisted of acetonitrile-0.1% phosphoric acid (25: 75, V/V) with a flow rate of lmL/min. The detection wavelength was 275 nm. Results:The linear range of baicalin in rabbit plasma was 0. 087 22 2.907 5 mg/L ( r = 0.989 9). Three relative recoveries of baicalin represented low, middle and high concentrations were 96.3% ,97.3% and 96.6% respectively. Conclusion: This method is accurate, reliable and simple and is suitable for the pharmacokinetic study of blood concentration of baicalin.
