HIF-1α在宫颈癌和宫颈上皮内瘤变中的表达及意义

摘要 为了探讨缺氧诱导因子-1α(HIF-1α)在宫颈癌及宫颈上皮内瘤变中的表达及生物学意义,利用免疫组织化学、Western blot和RT-PCR等方法检测了宫颈癌、宫颈上皮内瘤变、癌旁组织、正常组织和液基细胞中的HIF-1α的表达水平。研究结果表明,宫颈癌、CINⅡ-Ⅲ、CINⅠ和正常组织中HIF-1α的阳性表达率分别为58.9%(46/78)、48.0%(12/25)、6.67%(1/15)、0(0/12);宫颈癌组织中,HIF-1α表达与肿块大小、FIGO分期、组织学分级和淋巴结转移密切相关,与患者年龄无关;宫颈癌组织和CINⅡ-Ⅲ中的mRNA和蛋白水平均高于癌旁和正常组织及CINⅠ;液基细胞中宫颈癌和HSIL的mRNA和蛋白水平都高于LSIL及正常细胞。HIF-1α可能是宫颈癌形成过程中的早期分子事件,其表达增强与宫颈癌的浸润性发展密切相关,有可能成为新的早期筛查和判断宫颈癌生物学行为的诊断指标。 In order to investigate the expression and biological significance of hypoxia inducible factor-1 α (HIF-1α) in cervical carcinoma and cervical intraepithelial neoplasia, methods of immunohistochemistry, Western blot, RT-PCR and other methods were used to test the level of HIF-1α cervical carcinoma, cervical intraepithelial neoplasia, carcinoside tissue, normal tissue, and liquid-based cells. The results showed that the positive expression rates of HIF-1α in cervical carcinoma, CIN Ⅱ-Ⅲ, CIN Ⅰ, and normal tissue were 58.9% (46/78), 48.0% ( 12/25 ), 6.67% ( 1/ 15) , and 0% (0/12) respectively. In cervical carcinoma tissue, the expression of HIF-1α had close pertinence with the mass of carcinoma size, FIGO stages, histological grades, and metastasis of lymph node, but unrelated with the age of patients; the levels of mRNA and protein in cervical carcinoma tissue and CIN Ⅱ-Ⅲ were both higher than in carcino-side and normal tissues and CIN Ⅰ; the levels of mRNA and protein in liquid-based ceils and HSIL were all higher than in LSIL and normal cells. HIF-1α may be an early molecular event in cervical neoplasia, and its strengthening expression had close pertinence to infiltration development of cervical carcinoma, therefore, it may become an early diagnosis index to sieve check and judge the biological behavior of cervical carcinoma.
