摘要
研究HCCR在宫颈脱落细胞(来自宫颈液基细胞学残液)中的表达及其与HPV16/18感染的关系,探讨其对宫颈上皮内瘤变和宫颈癌诊断的意义。选取570例宫颈上皮内瘤变和宫颈癌液基细胞学残液标本中具有相应组织学标本者31例,并随机选取10例正常标本作对照,采用免疫组化法和原位杂交法检测HCCR和HPV16/18。结果可见,HCCR在正常宫颈细胞及组织中呈现低表达,在宫颈上皮内瘤变及宫颈癌细胞和组织中阳性率随着病变的升级而升高;HPV16/18 DNA的检出率随着宫颈病变的升级而升高;HCCR和HPV16/18在宫颈病变中的表达呈正相关(rs=1,P<0.005);无论是HCCR还是HPV16/18,在宫颈液基细胞学残液和相应宫颈组织中的检出率都是一致的。结论利用宫颈液基细胞学残液检测HCCR和HPV16/18对认识二者之间的关系及对宫颈上皮内瘤变和宫颈癌的筛查诊断具有重要意义。
The expression of human cervical cancer oncogene ( HCCR ) in cervical fall-off cells ( come from liquidbased cytological remant fluid) and its relationship to HPV 16/18 infection was studied in order to examine its significance to diagnose cervical intraepithelial neoplasia and cervial cancer. From 570 cases of cervical intraepithelial neo- plasia liquid-based cytological remant fluid samples 31 cases were chosen for those possessing corresponding histological samples; and another 10 cases of normal samples were chosen as control. Immunohistochemical method and hybridization in situ method were adopted to examine the HCCR and HPV 16/18. The results showed that the expression of HCCR appeared low in normal cervical cells and tissue, the positive rate of cervical intraepithelial neoplasia cells and tissue went high as the degree of pathology escalated; the detection rate of HPV 16/18 DNA also went high as the degree of cervical pathogy escalated; the expression of HCCR and HPV 16/18 in cervical pathology appeared as positive pertinence ( rs = 1, P 〈 0. 005 ) ; no matter HCCR or HPV 16/18, their detection rate was conformable in liquid- based cytological remant fluid and corresponding cervial tissue. Therefore, the use of liquid-bascd cytological remant fluid to examine HCCR and HPV 16/18 have great significance to know the relationship between both and to sieve check and diagnose cervical intraepithelial neoplasia and cervical cancer.
