摘要
大连化学物理研究所及其合作单位承担了多维液相色谱仪器研制与开发任务。研制了包括高压色谱恒流泵、紫外检测器、多维接口以及一体化控制软件的多维液相色谱仪,整个系统最高工作压力可达到40MPa,泵流量范围可以从1μL/min到9.999 mL/min;检测器可以在190~720 nm的波长范围内进行检测。将多维液相色谱仪应用于珠蛋白酶解产物的分析,离子交换色谱作为第一维,反相色谱作为第二维,整个系统的峰容量可以达到3306。
Dalian Institute of Chemical Physics, Chinese Academy of Sciences and its cooperation units took on the task of developing multi - dimensional liquid chromatography instrumentation. After several years' endeavor, high pressure constant -flow -rate pump, UV detector, multi -dimensional interfaces and integrated controlling software have been successfully developed. In this paper, the performance of the multi - dimensional liquid chromatography instrumentation was introduced in detail. The maximum operational pressure of the whole system was up to 40 MPa, the range of the flow rate was 1 μL/min - 9. 999 mL/min and the wavelength of UV detector was 190 - 720 nm. Finally, the performance of the multi -dimensional liquid chromatography instrumentation was demonstrated by the analysis of the digests of globin. The first dimension was ion exchange chromatography; the second dimension was reverse phase liquid chromatography. The peak capacity of the whole system was 3306.
