摘要
BCaCD_(885)细胞系是从一男性颊粘膜鳞状细胞癌患者原发灶所获得组织块,经体外培养2年已传180代以上。作者对原患者组织切片,体外培养不同代细胞和移植瘤用光镜、电镜进行观察,结果是三种细胞都具有鳞状上皮癌组织学特征。BCaCD_(885)细胞系的细胞生长快,失去接触性抑制,在软琼脂中形成克隆;其染色体众数为亚三倍体,畸变率高;在裸鼠体内具有成瘤性。BCaCD_(885)为已建成的恶性生物学特征稳定的确立细胞系。
A cell line designated as BCaCD885 has been established from a male with buccal mucesasquamous cell carcinoma and transfered for 180 passages within two years.Authors have observed morphology of the cell line with light and electron-microscope.It is showed deeply stained nuclei of various size and containing several nucleoli.Multinuc-leated giant cells with abnormal mitotic figures were seen.At the ultrastructure,the cellsarranged mass.There were many bundletonofibril in their cytoplasm,but it lacked ofsecretory granules.The cells were held together by desmosomes not junctional complexes.Contact inhibition in the cells was lost.The cells formed colonies on the soft agar.Thechromosome numbers of most of cells were subtriploid.The chromosome distortion ratewas high.Heterotransplantation showed that the tumor grew.The morphology of thetransplanted tumor were identical with that of the donor.Immunofluorescence of keratinwas positive in the transplanted tumor.
