摘要
目的:观察旋覆花素及苦碟子对过度训练大鼠肾组织细胞Bax和Bcl-2的表达以及Bax/Bcl-2比值变化的影响,探讨上述二种药物抗肾组织细胞凋亡的分子机制。方法:采用大鼠游泳至力竭建立过度训练模型,将大鼠随机分为安静对照组、力竭运动组(力竭6h、力竭24h)、旋覆花素防治组(旋覆花素6h、旋覆花素24h),苦碟子防治组(苦碟子6h、苦碟子24h)。采用免疫组织化学方法检测大鼠肾组织Bax、Bcl-2蛋白的表达;采用CMIAS病理图像分析仪测量Bax、Bcl-2蛋白表达的平均光密度。结果:力竭后6h及24h大鼠肾组织Bax的表达增强(P<0.05),Bcl-2的表达减弱(P<0.05),Bax/Bcl-2比值明显增高(P<0.05);旋覆花素和苦碟子6h及24h组大鼠与同期力竭组比较,Bax的表达减弱(P<0.05),Bcl-2的表达增高(P<0.05),Bax/Bcl-2比值降低(P<0.05)。结论:旋覆花素和苦碟子均可通过调节Bax、Bcl-2及Bax/Bcl-2比值而起到抗过度训练大鼠肾组织细胞凋亡的作用。
Objective:To understand the effects of Inula Britannica and Lxeris Sonchifolia Hance on the changes of expression of apoptosis regulatory genes Bcl 2 and Bax in kidney tissue in exhausted rats. Methods: The animal model of acute kidney in- jury induced by overtraining were developed by exhaustive swimming. 56 male wistar rats were randomly divided into sedentary control group, exhaustive swinmaing group( at 6 hour, 24 hour after exhaustive rat), Inula Britannica group (at 6 hour, 24 hour after exhaustive rat) and Lxeris Sonehifolia Fiance (at 6 hour,24 hour after exhaustive rat). The expression of Pax and Bcl 2 were tested by immunohistochemieal staining and CMIAS image analysis system. Results: The expression of Bax was stronger, the expression of Bcl - 2 was weaker, the Bax/Bcl - 2 ratio increased remarkably at 6 hour and 24 hour after exhaustive swimming. Alter treatment of Inula Britannica and Lxeris Sonchifolia Hance, compared with that of control rats, The expression of Bax decreaed remarkably, the expression of Bcl - 2 increased significantly, the Bax/Bcl - 2 ratio decreased remarkably. Conclusion: Inula Britannica and Lxeris Sonchifolia Hance could have the obvious anti apoptosis effect of renal cell by regulating the expression of Pax, Bcl 2 and the Pax/ Bcl - 2 ratio.
